How GPC/SEC Works
Dissolved gasses are removed from the mobile phase as the pump draws it through the degasser before sending it onwards to the autosampler and column oven. The autosampler draws sample from either a vial or a microtiter plate and injects it into the mobile phase which then continues to the column. As the dissolved sample flows through the porous column matrix, the individual molecules diffuse in and out of the pores. Since larger molecules are excluded from more pores, they will travel through the column faster and elute earlier while the smaller molecules elute later. The result is a separation by hydrodynamic size for measurement by the detectors.
